Description

FluoProbesŌ Fluorescent Protein Labeling kits are designed for the covalent labeling of proteins, enzymes or antibodies with molecular weights greater than 25 kD. The FluoProbes® kit uses a Succinimidyl ester dye to create a covalent bound with proteins. The conjugate is desalted with convenient Centrifuge Filter Columns.

Labeling and Purification

Each kit contains all components necessary for fluorescent labeling and purification, starting with enough reactive dye for up to five labeling reactions (each of up 1.5mg protein), buffers and desalting tools. Centrifuge Filter spin columns are provided for removal of unbound dye from labeled protein conjugates. A quick start optimized protocol allows easy labeling of 50 µg, 1mg of IgG or up to 1.5 mg protein. Guidelines are provided for changing the ratio of dye labels linked to a protein.

Applications

FluoProbes conjugates are widely useful for ELISA, flow cytometry, fluorescence microscopy, FRET, fluorescence polarization and a variety of fluorescent immunoassays.

FluoProbes 547 and 647 replace advantagely Cy3 and Cy5 labels, and can also be used for tandem applications. The 547H and 647H labels are improved hydrophilic versions of FP547 and FP647 labels, ideal for most protein labeling.
FluoProbes 488 replaces FITC, with higher brightness and unsurpassed photostability. For those who cant change FITC, choose FluoProbes495 which is fluorescein with improved spacer arm.

Workflow

The FluoProbes activated dye is mixed with protein, incubated for 45 minutes at room temperature and stopped for 30 minutes. The labeled protein conjugate is loaded onto provided centrifuge Filter and recovered via a 2-minute spin.

Typical Results

Greater than ninety percent recovery of dye labeled human IgG from a Centrifuge Filter spin column with a dye/protein ratio of 3 using the standard quick start kit protocol.
