Cell Lines:

Accutase has been shown effective on: primary fibroblasts, keratinocytes, vascular endothelial cells, hepatocytes, vascular smooth muscle cells, hepatocyte progenitors, primary chick embryo neuronal cells, bone marrow stem cells, adherent CHO and BHK cells, macrophages, 293 cells, L929 cells, immortalized mouse testicular germ cells, 3T3, Vero, COS, HeLa, NT2, MG63, M24 and A375 metastatic melanoma, gliomas U251 and D54, HT1080 fibrosarcoma cells, and Sf9 insect cells.

Application:

Accutase performs exceptionally well in detaching cells for: analysis of cell surface markers, virus growth assay, quiescence assays by serum starvation, transformation assays by oncogene transfection, neural crest cell migration assays, cell proliferation, cell haptotaxsis, tumor cell migration assays, routine cell passage, production scale-up (bioreactor), and flow cytometry.

Formulation:

Accutase has both protease and collagenolytic activity. Each lot of Accutase is individually formulated to meet quality control standards for effective cell detachment. Accutase is supplied in Dulbecco’s PBS containing 0.5mM EDTA·4Na and phenol red.

Availability:

Accutase is available in ready-to-use 100ml sterile bottles.

Cell Detachment

CELL DETACHMENT: Human MG63 fibrosarcoma cells cultured on tissue culture treated dishes in DMEM + 10% FBS were treated with ACCUTASE. Treatment results in rapid cell detachment, a single cell suspension, and high viability. ACCUTASE is gentle on cells; viability was 97 ± 3% even after 45 minutes in ACCUTASE.

Accutase has been developed to meet the most demanding needs for gentle and effective detachment of adherent cells, ad fibroblasts, endothelial cells, neurons, tumor cell lines, and insect cells.

Accutase gently and rapidly dissociates tissues for cell isolation and propagation. It is recommended for use with SmartPlastic adhesion cell culture plasticware

Accutase combines protease and collagenolytic activities which maximizes its versatility for cell detachment and tissue dissociation. It does not contain mammalian or bacterial-derived products.

Increase in cell

Count : Various constructs of genetically engineered CHO cells, BHK cells, and a hybridoma were grown in suspension in serum-free or protein-free medium. Representative cells aliquots were treated with an equal volume of PBS or ACCUMAX and incubated for 5 minutes at 37°C. Cell number was then determined with a Coulter Counter.

Accumax developed to meet the most demanding needs for an effective cell aggregate dissociation solution, has been proven effective in dissociating clumped cells in suspension cultures of hybridomas, CHO, BHK, 293, and others.

Accumax gently and rapidly dissociates cell clumps to yield single cell suspensions for accurate and reproducible cell counts.
