Protein w. FluoProbes647H but nothing w. flow cytometry

Hello,

A colleague labeled a protein with FluoProbes647H but didnt detect anything with a 660nm filter by flow cytometry.
I'm confused because she says the conjugate appears to be blue in color visually... Is it possible? What is the visual color of the deliverd dye in solution (before labeling) and once conjugated?
If blue appearance was possible, I could suspect rather there is a problem in FCM detection settings, because it seems she is not used to detect in this color.
If blue is impossible, I fear a wrong fluorophore was used...

Thanks !

!hello Azar,

The FP-647H is blue when dissolved in water.
The general problem when communicating about colours of dyes that we have to distinguish between absorption and emission colour.
The FP-647H is blue by absorption and emits far red fluorescence light.

Hope it could help you.
