DAB Peroxidase Substrate System - Code E885

Directions for Use

Contents:

DAB Substrate Tablets (Code E733)

Peroxidase Solution (Code E882)

Mixing/Dispensing Bottle

Each Tablet will yield 10 ml of substrate solution, enough to effectively detect 50cm2 of membrane or about 20 slides.

Step 1: Drop one tablet into the mixing/dispensing bottle, and add 10 ml deionized water. Cap bottle and shake gently until the tablet dissolves, about 2-3 minutes.

Step 2: Using the attached dropper cap, add one drop of peroxidase solution to the dissolved substrate tablet.  (Caution: Do not use the dropper cap from the mixing bottle to aliquot the peroxidase solution.  Doing so may contaminate the peroxidase solution and decrease the shelf-life of the reagent.)

Step 3: Cap the mixing bottle and invert gently to mix.

Step 4: Using the dispensing dropper, layer the prepared substrate solution onto  the membrane or slide.  Generally, 1 ml of substrate will be sufficient for detecting 5-10 cm2 of membrane.  0.5 ml of substrate is sufficient for most slides.

Step 5: Unused Substrate Solution can be stored at 4C (refrigerated) for up to 6 hours.
