A new compact, convenient solid gel form of 50X TBE!

 

While tackling the issue of precipitate forming in 10X TBE stock solutions, AMRESCO’s R&D staff came across the idea of an alternative form of the buffer. This would be a buffer that could withstand prolonged periods of storage and deliver optimal performance upon use. Knowing that today’s sophisticated electrophoresis techniques require high-quality reagents at optimal, consistent concentrations, a new solid-gel state of TBE in a 50X concentration has been developed.

A liquid state of Easy TBE 50X Concentrate is cast at a temperature range which insures the uniformity of all components. Only AMRESCO’s highest purity Tris, Boric Acid, and EDTA (disodium salt) are used. Each lot of material is tested for proper pH and conductivity specifications.

The solid-gel tablets are packaged in easy-to-open pouches. You just tear the pouch, place the tablet into a 250 or 500 ml microwavable beaker, and pipette

10-20 ml of distilled water into the beaker. After microwaving the tablet for 20-25 seconds, remove the beaker from the microwave (Be careful!), add 10-20 ml of distilled water and pour the newly liquefied TBE into the proper sized graduated cylinder. Rinse the beaker with distilled water and pour into the graduated cylinder. Bring up the TBE to the correct final volume and GO!

AMRESCO offers two sizes of Easy TBE 50X Concentrate for your convenience. The 10 ml tablet results in 500 ml of 1X TBE; the 20 ml tablet results in 1 Liter of 1X TBE. Several package sizes are available.

 

Procedure:

Heating Guidelines

  • Due to Microwave oven variances, times may vary according to model/brand used. The following instructions were developed using a 400 watt microwave.

  • Remove the tablet from the pouch by simply tearing where indicated by the notch on either side of the pouch.
  • Place the tablet into a 250-500 ml beaker.
  • Introduce approximately 10 ml of distilled water into the beaker via pipetting or squirt bottle. NOTE: This step must always be performed. The addition of the water eliminates any risk of eruption when heating the solid material.
  • Place the beaker into the microwave.
  • Microwave on high for 5-10 seconds (10 ml tablet) or 20-25 seconds (20 ml tablet). Times may need to be adjusted according to the model/brand of microwave used.
  • Using heat-resistant gloves, remove the beaker from the microwave and gently swirl. The material should now be in a liquid phase. If not, re-microwave in 5 second intervals until material is in a liquid phase. Again, it is crucial to have approximately 10 ml of distilled water in the beaker.
  • Next pour the liquid TBE concentrate into an appropriately sized graduated cylinder.
  • Rinse the beaker that was used during the heating process with distilled water to recover any remaining TBE concentrate. Transfer this rinse to the graduated cylinder that contains the TBE concentrate.
  • Add additional distilled water until desired final volume is achieved.
