Directions for use

EZ Squeeze Gene-PAGE PLUS (PAGE-PLUS blended with urea in a single-use, disposable bottle) features AMRESCO’s blend of modified acrylamide and a proprietary cross-linker to form a superior matrix in nucleic acid electrophoresis. The resulting unique pore structure in PAGE-PLUS gels leads to improved resolution, running time, and band spacing over that of conventional acrylamide:bis-acrylamide blends. In addition, post-electrophoretic fixation is no longer necessary, making PAGE-PLUS products the ultimate matrices for your electrophoresis needs.

Gene-PAGE PLUS is available in several different ready-to-use concentrations specifically formulated for nucleic acid sequencing (no filtering is necessary).Gene-PAGE PLUS gels can be stained and processed according to standard post-electrophoretic methods while

exhibiting little to no background, making PAGE-PLUS the superior replacement for acrylamide:bis-acrylamide mixtures in electrophoresis of nucleotides. Use of these specialized blends result in at least 40% more sequence per gel without compromise of integrity. In fact, Applied Biosystems Inc. reports obtaining "read lengths of 800 bases or more...,"** when using PAGE-PLUS on their ABI Prism 377 DNA Sequencer.

What could be easier? With urea already blended in the solution (Gene-PAGE PLUS) and stabilities of 6 months, the addition of the unique solid phase TEMED and ammonium persulfate disks are all that is required of the investigator. Suggested Parameters:

(As with any material, some experimentation may need to be performed to determine the optimal formulation for individual use).

 

Manual Sequencing:

  • For best results, this product should be allowed to warm to room temperature before use.

  • Add the Ammonium Persulfate TEMED polymerization catalyst/initiator disks. Mix the solutions by gentle inversion for 2-3 minutes (to avoid bubbles) and immediately pour into a prepared glass plate assembly. Insert an appropriately sized comb into assembly and allow to polymerize completely. (Generally, waiting 1 hour before beginning electrophoresis leads to optimum results). PAGE-PLUS gels do not require post-electrophoretic fixation. (For manual sequencing, it is best to keep the running temperature at 51°C or below).

Automated Sequencing:

Again, depending on instrumentation, chemistry utilized, and aim of read, some experimentation may need to be done to optimize the conditions for your specific parameters.
