Directions for use


Reagents Provided:
  • Component 1 Stacking Gel Mix

  • Component 2 Resolving Gel Buffer

  • Component 3 Resolving Gel Concentrate

 

Reagents Required (Not provided in this package)
  • Ammonium Persulfate (10% solution, freshly made) (code 0486)

  • TEMED (code 0761)

  • Water-saturated Butanol (optional)

Add the entire contents of the bottle of Component 3 (Resolving Gel Concentrate) to the bottle of Component 2 (Resolving Gel Buffer) and mix gently. This mixture is stable when stored at 4°C for up to three months.

 

For the resolving gel:

Aliquot the desired amount of Pro-PAGE mix into a sidearm flask, stopper and degas under gentle vacuum for ten minutes, or until bubbles no longer form on the surface of the liquid.

For each 10 ml of degassed resolving gel mix, add 100 L of 10% Ammonium Persulfate solution and 10 L of TEMED. Mix by gently swirling the flask, and immédiatement pour into a prepared glass plate assembly. If a stacking gel will be used, overlay the resolving gel with water-saturated butanol, if desired, and allow to polymerize fully (about 30 minutes). If no stacking gel will be used, insert the appropriate comb and allow to polymerize fully.

 

To prepare a stacking gel:

When polymerization of resolving gel is complete, remove the water-saturated butanol by aspiration, and prepare the stacking gel as outlined below.

Aliquot the desired volume of Component 1 (Stacking gel mix) into a sidearm flask, and degas as above. For each 10 ml, add 100 L of 10% Ammonium Persulfate and 10 L of TEMED. Mix by gently swirling and immediately pour this mixture on top of the resolving gel. Insert appropriate comb, and allow to polymerize fully (30 minutes).
