Directions for use

Thermo-PAGE is a novel acrylamide blend designed for use in sequencing applications that require high voltage, shorter runs. These faster runs combined with the high resolution provided by Thermo-PAGE generate more data in less time.

Specifically, Thermo-PAGE has been shown to be highly effective when used on the ABI 377 4X modules for 3.5 hour runs at 3000V. At 5.0% acrylamide solution, 6M urea and 1X TBE, Thermo-PAGE has been shown to generate as many as 700bp read lengths when using the described run conditions.

Thermo-PAGE is available as a 40% concentrate or as a convenient 5% premix solution which contains 6M urea and 1X TBE. Handling and use of Thermo-PAGE for sequencing applications is the same as standard acrylamide solutions. The concentrate can be diluted to any desired concentration to optimize for individual applications. Thermo-PAGE 5.0% premix has been determined to be effective under the running conditions stated above. The pre-mix provides the added convenience of simply adding Ammonium Persulfate and TEMED at standard concentrations and pouring the gel.
