Directions for Use

 

For Most DNA Applications:

 

  • Add the entire contents of the small bottle of buffer to the large bottle of Phenol:Chloroform and gently mix.

  • Allow the layers to separate before use, approximately 2 to 4 hours.

  • The pH of the Phenol:Chloroform solution is now 8.0 + 0.2.

  • After addition of the buffer, the solution is stable for more than six months when stored at 4C.

 

For DNA and RNA Applications where a lower pH is more suitable:

  • The large bottle of Phenol:Chloroform should be used directly with no additions.

  • The pH of the large bottle of Phenol:Chloroform is 6.7 + 0.2 and is stable for one year when stored at 4C.

 

For Purification of RNA at an acid pH:

  • Try AMRESCO's 5:1Phenol:Chloroform, pH 4.5 (code E277) or 1:1Phenol:Chloroform, pH 5.2 (code 0966) both complete with Isoamyl Alcohol.
